5-Ethynylcytidine is a fluorescent analog of cytosine that has been synthesized for use in biochemical research. It is used to identify cellular sites of RNA synthesis and replication, as well as to study the role of the 5-ethynyl group in the mechanism of DNA methylation. The synthetic route starts with an azide intermediate which is then treated with sodium hydroxide, followed by hydrolysis and demethylation. 5-Ethynylcytidine has been shown to be selective for mammalian cells and tissues, and was incubated with calf thymus DNA for crystallographic data.
