RGB-286638 is a non-competitive inhibitor of the enzyme DNA topoisomerase II. It has minimal toxicity and a low potential for drug resistance. This agent has been shown to have potent antitumor activity in mice with cancer, inhibit cell growth in cultured cells, and cause necrotic cell death. RGB-286638 binds to the MCL-1 protein and inhibits the production of cytokines that are important for regulating immune responses, such as tumor necrosis factor (TNF). This agent also has a high affinity for actin filaments, which may be due to its ability to bind to myosin heavy chains.
