16α-Hydroxytestosterone is a metabolite of testosterone that is formed from the oxidation of this hormone by cytochrome P450 enzymes. It has been shown to inhibit the activity of various enzymes, such as NADPH-cytochrome P450 reductase, NADH-cytochrome P450 reductase, and CD-1 mice liver microsomal epoxide hydrolases. This metabolite has been found in experimental models to cause cancerous tumors in rats, and it can also be used to generate monoclonal antibodies against these enzymes. 16α-Hydroxytestosterone binds reversibly to enzymes such as NADP+-dependent dehydrogenases and hydroxylases. The reaction mechanism for this binding involves a dinucleotide phosphate intermediate and anhydrous sodium.
