Coelenterazine H is a cell-permeable, photoreactive molecule that has been used in the detection of ATP levels. The effects of coelenterazine H on ATP levels are dependent on its activation by light. The luminescence signal is proportional to the amount of ATP present, and can be measured using a luminometer or spectrophotometer. Coelenterazine H binds to effector proteins in the cytoplasm and triggers intracellular signaling pathways, which leads to an increase in cytosolic calcium and activation of luciferase. This process can be activated with low light and is often used in cell culture to measure ATP levels. Coelenterazine H also has protease activity, which may degrade this molecule after it has been activated by light. The bioluminescence signal is dependent on luciferases present in the cell and their ability to produce light when they are active. This process requires ATP as well as a molecule called camp.,
