DNA damage-binding protein 2 is able to recognise and bind to UV-induced DNA lesions and facilitates efficient recognition by XPC. DDB2 and XPC then initiate global genome nucleotide excision repair (GG-NER) to protect DNA from mutagenesis associated with premature aging and skin cancer. Timely DDB2 dissociation is required for DNA damage handover to XPC and swift progression of the multi-step repair reaction. Damage handover from DDB2 to XPC coincides with the arrival of the TFIIH complex, which further promotes DDB2 dissociation and formation of a stable XPC-TFIIH damage verification complex. DDB2 binds directly to and flips out UV-induced damaged bases to create a more suitable substrate for XPC.The UV-DDB complex is part of a larger E3 ubiquitin-ligase complex (CRL4DDB2), also containing CUL4A, RBX1, and the COP9 signalosome. AF647 dye is a commonly used bright, far-red-fluorescent dye which is pH-insensitive over a wide molar range.
