APOA4 (135-143) Heavy is derived from the APOA4 protein, a component of the apolipoproteins which make up High-density lipoproteins along with lipids and enzymes. Synthesised in the intestines as a preproapolipoprotein, APOA4 is a 46kDa glycoprotein composed of amphipathic alpha-helices comprised of 22-mer amino acid repeats. Functionally APOA4 is involved in lecithin cholesterol acyltransferase activation and Cholesteryl ester transfer protein activity. It plays a further role in carbohydrate metabolism through increasing insulin secretion and hence improving glucose homeostasis. It also regulates the nuclear receptor NR1D1, resulting in the inhibition of hepatic gluconeogenesis, and functions as an anti-oxidant through the prevention of lipoprotein peroxidation.APOA4 has a low affinity for plasma lipoproteins, thus it takes a lipid free form when circulating in the blood. Moreover the plasma level of APOA4 is reduced in coronary artery patients compared to healthy controls.The leucine residue at position 8 has been isotopically labelled with carbon-13 (6) and nitrogen-15 (1), giving this peptide a mass increase of 7 compared to the unlabelled peptide.
