1-(2-Pyridyl)-1H-indole is an alkynyl compound that has been synthesized by a regioselective Friedel-Crafts reaction. The intramolecular cyclobutanol formation is the rate-determining step of this transformation. This reaction was found to be highly efficient and scalable for the synthesis of 1-(2-pyridyl)-1H-indoles in good yields. The alkenylation of these compounds with acetylene derivatives was also shown to be an efficient route to access different functionalized 1-(2-pyridyl)-1H-indoles.
