Actein is a biologically active compound extracted from the root of Cimicifuga foetida. It has been shown to have cytotoxic effects on human breast cancer cells and human leukemia cells. Actein induces apoptosis through activation of the mitochondrial pathway by binding to the Bcl-2 family protein, Bax. Actein also has an effect on mitochondria membrane potential, which may be due to its interaction with K+ channels in the outer mitochondrial membrane, leading to increased permeability and more leakage of K+ ions into the cytosol. This leads to depolarization of mitochondria and subsequent activation of caspases. Actein also inhibits autophagy by inhibiting lysosomal proteolysis.
