1,3,5-Trimethylpyrimidine-2,4(1H,3H)-dione is a reactive compound that can undergo oxidation to form covalent bonds with DNA. It is soluble in water and has a low first-order rate constant for its reaction with amines. The second-order rate constants for reactions of 1,3,5-trimethylpyrimidine-2,4(1H,3H)-dione with DNA bases have been determined using an analytical HPLC method. This compound was found to be an effective inhibitor of the oxidative DNA damage induced by caffeine in human cells. The redox potentials of this compound and its derivatives are very close to those of hydrogen peroxide and the carbonyl group on the molecule can activate hydrogen peroxide. The activation energies for these reactions have been determined experimentally and are available in the literature.
