2'-Amino-[1,1'-binaphthalen]-2-ol (BINOL) is a chiral enantiopure amine that is used as a ligand for asymmetric synthesis. BINOL can be prepared through a cross-coupling reaction of an N-trifluoromethanesulfonyloxy acetyl bromide and an α-aminonitrile. The resulting product is then hydrolyzed to the corresponding amine, which can be purified by recrystallization. BINOL is often used as a ligand in asymmetric hydrogenation reactions with acids such as picolinic acid or trifluoroacetic acid. In these reactions, the hydrogen atom has a higher affinity for the protonated form of the ligand than for the neutral form, leading to an increased rate of reaction on one side of the molecule.
