4-Aminohexanoic acid is a non-proteinogenic amino acid that is found in wheat germ lectin. It has been shown to be an excellent substrate for the enzyme urokinase-type plasminogen activator, which cleaves 4-aminohexanoic acid into 4-aminoheptanoic acid and ammonia. The reaction mechanism of this enzyme is not well understood, but it is thought to involve the hydrolysis of a phosphoramide bond. This hydrolysis produces a nucleophile that attacks the carbon at the alpha position of the amine group on 4-aminohexanoic acid. A carbocation intermediate is formed, which can then react with chloride ions to form an amide bond between the two carbons on the amino acid backbone. This mechanism may also explain how other enzymes activate 4-aminohexanoic acid using different mechanisms.
