Piperazine-1-carboxylic acid amide is a non-opioid agent that prevents the release of histamine from mast cells by blocking the IL-2 receptor. It has been shown to be effective in treating chronic cough, and provides relief within hours of treatment. Piperazine-1-carboxylic acid amide also has an inhibitory effect on TRPM8 channels, which are involved in sensing cold temperature and pain sensation. This drug is metabolized through a number of metabolic transformations including hydrolysis by esterases or glucuronidases, oxidation by cytochrome P450 enzymes, reduction by glutathione reductase, or conjugation with glucuronic acid. The pharmacokinetic properties of piperazine-1-carboxylic acid amide have not been fully characterized.
