4-Benzyloxygramine is an alkaloid that is a benzyl derivative of the indole nucleus. It has been shown to be an antagonist of lithium, calcium and potassium channels. The synthesis of 4-benzyloxyindole from 2-methyl-3-nitrophenol was achieved through a convergent approach. The debenzylated product was hydrolyzed with sodium hydroxide in water, followed by decarboxylation with aluminium chloride to form the desired product. 4-Benzyloxygramine reacts with benzaldehyde in the presence of potassium hydroxide to form 4-benzyloxyindole. This reaction involves an elimination of hydrogen bromide and formation of a double bond between carbon atoms 3 and 4 on the indole ring.
