5-Bromo-2,4-diamino-6-hydroxypyrimidine (5BDAP) is a ribonucleotide and aminoimidazole analog that has been shown to be a potent inhibitor of the enzyme dihydrofolate reductase. 5BDAP is structurally similar to the natural substrate for this enzyme, thymine, and like other analogues, it competes with the natural substrate for binding to the enzyme's active site. 5BDAP binds to the enzyme and blocks its activity, preventing it from reducing dihydrofolic acid into tetrahydrofolic acid. This prevents the production of thymidylate synthase and thymidylate, which are key enzymes in DNA synthesis.
