Description

4-Chloro-3-chlorosulfonylbenzoic acid is a potent inhibitor of the shikimate pathway and has been shown to be an anti-tuberculosis agent. The compound has a chloro substituent at the 4 position, which allows it to act as an electron-withdrawing group that stabilizes the transition state. This substitution also increases the activity by 10 times, providing an explanation for its potency against tuberculosis. The active site of 4-chloro-3-chlorosulfonylbenzoic acid is located in the chorismate binding pocket of shikimate dehydrogenase, where it acts as a competitive inhibitor. The crystal structure of this inhibitor bound to shikimate dehydrogenase has been determined by x-ray crystallography and shows that the lead compound binds in a manner similar to other inhibitors with high affinity.
