Coumarin suberoylanilide hydroxamic acid (CSHA) is a hydroxamic compound that is used in cancer research. It inhibits the activity of histone deacetylases, which are enzymes that remove acetyl groups from lysine residues on histones and other proteins. CSHA has been shown to inhibit the growth of tumor cells by decreasing the expression of genes involved in energy transfer, DNA synthesis, and RNA synthesis. The fluorescent probe CSHA can be used to study the localization of these enzymes in cells. Coumarin-saha is a derivative of CSHA that has been modified to have an affinity for cysteine residues found in proteins such as histones and RACK1. Coumarin-saha has also been shown to inhibit the activity of some enzymes involved in DNA replication such as DNA polymerase alpha and beta.
