N-Cbz-glycine 4-nitrophenyl ester is a substrate for serine proteases. It is used to study the reaction mechanism of serine proteases and the rate of hydrolysis at different pH values. N-Cbz-glycine 4-nitrophenyl ester binds to the active site serine residue in a protonated form, and it is released from the enzyme by an acid or base. The binding constants are determined by measuring the change in absorbance of the nitrophenolate moiety at 340 nm with respect to time. This synthetic substrate is used as a reference compound for measuring catalysis by serine protease enzymes and ionization rates during hydrolysis reactions.
