Eflornithine is a drug that inhibits polyamine synthesis. It has been used in combination with nonsteroidal anti-inflammatory drugs for the treatment of cardiac arrhythmias, angina pectoris, and congestive heart failure. Eflornithine hydrochloride is also an inhibitor of DNA topoisomerases I and II. These enzymes are important in the regulation of DNA replication and transcription. The inhibition of these enzymes leads to cell death by inhibiting phosphorylation reactions required for the production of DNA. Eflornithine hydrochloride has minimal toxicity and is not known to cause toxic effects on the heart, lungs or kidneys.
