Etodolac is a nonsteroidal anti-inflammatory drug (NSAID) that is used to relieve pain, inflammation, and stiffness. It is a racemic mixture of the enantiomers (+) and (-)-etodolac. Etodolac is a prodrug that can be activated by esterases to produce the active form, which inhibits cyclooxygenase (COX). COX catalyzes the conversion of arachidonic acid to prostaglandin H2, one of the mediators of inflammation. Etodolac has been shown to be effective in reducing symptoms of chronic arthritis and postoperative ileus. The drug acts on inflammatory cells and causes vasoconstriction. Etodolac's mechanism of action may involve alkylation at the level of DNA or protein synthesis, as well as its ability to inhibit prostaglandin synthesis. This drug also has an analgesic effect mediated by its ability to block transmission in central neurons
