Description

4-Thiothymidine is a nucleoside analog that is used in the treatment of cancer. It binds to the phosphoribosyl transferase enzyme, which is necessary for the synthesis of thymidylate from uridine. This prevents DNA replication and leads to cell death. 4-Thiothymidine can also be used in combination with other drugs such as l1210 and uridine. The drug binds to an allosteric site on uridine phosphorylase, which is an enzyme that is vital for the conversion of uridine into thymine. The binding inhibits nucleophilic attack by phosphate on the carbonyl group at position 3 of the substrate, preventing phosphorylase activity. 4-Thiothymidine has been shown to increase muscle mass in animal models.
