2-fluoro-5-iodobenzonitrile is a potent inhibitor of GSK3β, which is an enzyme that has been implicated in the regulation of cell proliferation, differentiation and apoptosis. 2-fluoro-5-iodobenzonitrile inhibits the activity of GSK3β by binding to its ATP binding site and blocking the formation of phospho-GSK3β. This inhibition leads to increased levels of cyclin D1 and caspase 3 activities, as well as reduced levels of Bcl2 protein. The kinetics of 2-fluoro-5-iodobenzonitrile have been studied in vitro using assays for kinases, cycloadditions, riboside and imidazolide. The orientation of 2-fluoro-5-iodobenzonitrile has also been determined using XANES measurements on rock2 crystals. These crystals were found to be oriented at 94° with respect to
