Z-gly-Pro-OH is a synthetic peptide substrate that has been used to study the activity of serine proteases, specifically histidine-specific proteases. It has been shown to be hydrolyzed by human liver and intestinal enzymes, including serine proteases, in vitro. This peptide also reacts with epirubicin, a drug that is used in chemotherapy for treating cancer. The reaction between the two compounds results in an increased potency of epirubicin and a decreased potency of z-gly-pro-OH. Z-gly-Pro-OH can be synthesized from L-histidine and glycylproline using the method of intramolecular hydrogenation to form an amide bond between the amino acid and the proline residue. The synthesis can be completed using a chromatographic method or titration method.
