Peroxynitrite is a reactive oxygen species that can cause cellular damage. It has been shown to be synthesized by the reaction of nitric oxide with superoxide, and is an important mediator of cell damage. Pre-incubation of cells with peroxynitrite leads to the modification of proteins in the extracellular environment, which causes a loss of function. The 6-hydroxyl group then reacts with amines and leads to the formation of nitrosamines which are cytotoxic. Peroxynitrite also inhibits RNA synthesis and DNA repair, leading to cell death. Peroxynitrite is cytotoxic to Mcl-7 cells and other cell lines, as well as myeloid leukemia cells (HL-60).
