4-Hydroxy-7-methoxycoumarin (4OHM) is a fluorescent compound that is used as a pharmacological probe. It has been shown to activate the mitogen-activated protein kinase (MAPK) cascade, which is involved in regulating cell growth and proliferation. 4OHM also induces the release of interleukin (IL)-1β and prostaglandin E2 from human monocytes, which are involved in inflammatory diseases. 4OHM has also been shown to induce necrosis in cells with low levels of extracellular ATP by inhibiting the enzyme phosphatase 2A. This inhibition leads to increased levels of intracellular calcium ions and reactive oxygen species, leading to necrosis through activation of proteases such as calpain.
