4-Methyl-7H-pyrrolo[2,3-d]pyrimidine is an anti-HIV drug that has been shown to have a potent anti-HCV activity in molecular modeling studies. The compound has been optimized for binding to the HCV NS5B RNA polymerase and the lead discovery step has been completed. 4-Methyl-7H-pyrrolo[2,3-d]pyrimidine is a ligand that binds to the C2' hydroxyl group of the nucleoside analogs and prevents them from being incorporated into the growing DNA chain. This leads to a halt in viral replication. The conformation of this molecule was determined by molecular modeling techniques and its amino group was bioisosterically replaced by a carbocyclic nucleoside.
