3,4,5,6-Tetrahydro-5-methyl-1-phenyl-1H-2,5-benzoxazocine hydrochloride (THPP) is a drug that is used as an analgesic and anti-inflammatory agent. THPP interacts with nonsteroidal anti-inflammatory drugs (NSAIDs) to cause neuronal death. It has been shown to inhibit the plate test for prostaglandin synthesis in human erythrocytes. The disulfide bond between cysteine residues on the protein molecule is susceptible to oxidative cleavage by reactive oxygen species from reactive oxygen species such as hydrogen peroxide and lipid peroxidation products. This can lead to the formation of a thiolate ion and a sulfenic acid intermediate. THPP is also known to produce chronic oral toxicity in rats at doses of 837 mg/kg/day for two months.
