3-Nitropyrrole is a synthetic compound that has been shown to have significant cytotoxicity in cell cultures. It is a nucleobase analog and can be detected by acid detection methods. 3-Nitropyrrole is an analog of pyrrole, which contains a nitro group on the third carbon atom. The nitro group confers significant cytotoxicity to cells, as well as mismatches in DNA sequences when used in polymerase chain reactions (PCR). 3-Nitropyrrole can be synthesized by reacting 2-amino-5-nitrothiophene with chloroform. Nitro groups are also found on the second and fourth positions of the molecule.
3-Nitropyrrole has been shown to inhibit DNA duplex formation and hydrogen bonding, which may be due to its ability to interact with primers or dNTPs. This interaction prevents hybridization of the two strands of DNA.
