Nodakenitin is a polymerase chain inhibitor that binds to the active site of the bacterial DNA polymerase and prevents the extension of DNA. Nodakenitin has shown potent activity against oral pathogens, including Streptococcus mutans, which causes dental caries. Nodakenitin is effective at inhibiting the growth of these bacteria by preventing the synthesis of new DNA (during which time the cells commit suicide). The matrix effect may be responsible for nodakenitin's effectiveness against oral pathogens. Nodakenitin also inhibits mitochondrial membrane potential and induces apoptosis in HL-60 cells.
