Alkaline phosphatase is a protein that is normally found in the cells of the liver, bile ducts, and pancreas. It plays an important role in the digestion of dietary fats and proteins by catalyzing the hydrolysis of phosphoric acid esters. Alkaline phosphatase has been used as a marker for udca therapy, which is a drug treatment for chronic liver disease. It has also been shown to have biochemical properties that are useful in transfection experiments with mammalian cells. The enzyme binds to p-nitrophenyl phosphate and cleaves it into p-nitrophenol and free phosphate ions. This reaction can be measured spectrophotometrically at 405 nm. Alkaline phosphatase has also been shown to have a signal peptide sequence that directs it to the endoplasmic reticulum membrane where it interacts with toll-like receptor 4 (TLR4).
