Phorbol esters are nonsteroidal anti-inflammatory drugs that also have potent inducers of protein kinase. They bind to the protein receptor, which is a transmembrane glycoprotein, and this binding induces phosphorylation of the receptor. The phosphorylated receptor then binds to DNA-dependent RNA polymerase, thereby inhibiting transcription and replication. The potent inducers of protein kinase include phorbol, phorbol esters, and other related compounds. Phorbols can be synthesized from diterpenes by reacting them with an alcohol in the presence of a strong base like sodium hydroxide or potassium hydroxide. This reaction produces a mixture of dihydrocompounds called phorbols. The model system for studying the biological activity of these compounds is HL-60 cells because they are sensitive to induction by these compounds and also produce chemiluminescence when exposed to them.
