2-Thiophenesulfonyl chloride is a nucleophilic compound that reacts with electron rich groups such as hydroxyl and amino groups. It has shown anticancer activity in animal models and is being studied for the treatment of bowel disease, autoimmune diseases, and inflammatory bowel disease. 2-Thiophenesulfonyl chloride inhibits serine proteases by reacting with the active site cysteine or histidine residue. This reaction produces an acyl enzyme intermediate which can be hydrolyzed to regenerate the original enzyme or react with another molecule of 2-thiophenesulfonyl chloride to form an irreversible covalent bond. 2-Thiophenesulfonyl chloride also inhibits the ns3 protease and macrocyclic inhibitors, which are enzymes involved in inflammation and cancer metastasis.
