1,3,6-Trimethyl-5-iodouracil (TMIU) is a nucleophile that reacts with electrophiles. It is used as a precursor to the synthesis of other compounds, such as 1,3,6-trimethyluracil. TMIU can be synthesized by chlorinating 6-methyluracil in the presence of a proton source. The reaction time for this synthesis is typically between 30 minutes and 3 hours. The yields are typically around 50%. Carbocations that form during this reaction are an analogous to methyl cations in nucleophilic substitutions. Bromination of TMIU has also been shown to produce bromo derivatives.
