Thieno[2,3-b]pyridine is a quinoline derivative that has been shown to be a potent antitumor agent. It inhibits the growth of human liver cancer cells and human breast cancer cells. Thieno[2,3-b]pyridine binds to the Pim-1 protein, which regulates cell proliferation. This interaction blocks the activation of cyclin D1 and inhibits the production of downstream proteins such as c-Myc, p27kip1, and cyclin E. Thieno[2,3-b]pyridine also inhibits the activity of DNA topoisomerase I by binding to its active site. The inhibition leads to accumulation of DNA breaks in tumor cells that are not repaired by this enzyme.
