D-Tyrosinol is a tyrosine analogue that is used in the treatment of chronic lymphocytic leukemia. It inhibits the enzyme adenine phosphoribosyltransferase, which is involved in the synthesis of purines and pyrimidines. The binding experiments on mammalian cells revealed that D-tyrosinol binds to the active site of adenine phosphoribosyltransferase (APRT) with high affinity, as well as inhibiting the enzymatic activity. A study on anticancer activity showed that D-tyrosinol inhibited leukemia cells by inducing apoptosis but not normal cells. The mechanism studies confirmed that D-tyrosinol binds to APRT and blocks its catalytic site, thereby inhibiting its activity. This inhibition leads to an accumulation of adenosine diphosphate (ADP) ribose and subsequent activation of protein kinase C (PKC). In addition, PKC interacts with mitotic checkpoint
