3-(Trifluoromethyl)benzamide is an acyl halide that is a potent, selective, and orally bioavailable b-raf inhibitor. 3-(Trifluoromethyl)benzamide has been shown to inhibit the growth of colon cancer cells in vitro and in vivo. This compound also has the ability to cross the blood-brain barrier, which may be due to its high lipophilicity and low molecular weight. 3-(Trifluoromethyl)benzamide inhibits b-raf phosphorylation by blocking the cb2 receptor agonist binding site on b-raf. This inhibition results in decreased levels of c-jun N-terminal kinase (JNK), which leads to apoptosis. The pharmacophore for this drug includes a hydrophobic region that binds to the ATP binding site of b-raf and a carboxylic acid moiety that binds to the serine/threonine
