Description

10-Hydroxy-16-epiaffinine is a natural product that was isolated from the hexacyclic monoterpenoid indole alkaloid, Rauvolfia tetraphylla. It has cytotoxic properties and has been shown to inhibit the proliferation of human lung carcinoma cells (HL-60) and murine leukemia cells (MFC-7). 10-Hydoxy-16-epiaffinine also inhibits the production of tumor necrosis factor alpha, interleukins 1 beta, and 6, and nitric oxide in HL-60 cells. The molecular modeling study revealed that the compound binds to DNA at two sites: one site near the junction between the major groove and minor groove and one site opposite to it. This binding leads to inhibition of DNA synthesis by preventing the unwinding of DNA strands.
