4-Fluoro-N-methyl-N-[4-[6-[(1-methylethyl)amino]-4-pyrimidinyl]-2-thiazolyl]benzamide is an allosteric modulator of metabotropic glutamate receptors. It has been shown to inhibit lipid biosynthesis in cells (e.g., COS cells) and has been used as a radiation sensitizer for low energy photon therapy for cancer treatment. 4FMTAB binds to the extracellular signal domain of the metabotropic glutamate receptor, which is a G protein coupled receptor. This binding inhibits the signal detection process, leading to a decrease in intracellular cAMP levels and subsequent inhibition of protein synthesis. 4FMTAB also inhibits translation by binding to the ribosome and inhibiting peptide bond formation between amino acids.
