Pemetrexed disodium is a drug that belongs to the group p2 of compounds. It is used in the treatment of colorectal cancer and other solid tumours such as lung, breast, stomach, and prostate cancers. Pemetrexed disodium is a prodrug which undergoes activation by hydrolysis to pemetrexed. Pemetrexed disodium has been shown to inhibit the production of thymidylate, which is an essential component for DNA synthesis. The minimal toxicity of this drug is due to its low reactivity with plasma proteins and lack of enzyme induction. Pemetrexed disodium has been shown to cause mitochondrial membrane potential loss in cells treated with radiation therapy. This loss activates apoptosis-inducing factor (AIF) and caspase-mediated cell death pathways.
