Guajadial is a natural product that belongs to the family of guaianolides. This compound has been found in an acetate extract of the bark of the South American tree Guajacum sanctum. Guajadial has shown cytotoxic activity against cancer cells and is being studied for its use as a chemotherapeutic agent. The antitumor activity of guajadial has been shown to be related to its ability to inhibit endothelial cell proliferation, which is essential for tumor growth and metastasis. A number of clinical studies have been conducted with this drug in patients with various types of cancer, including breast and prostate cancers. The mechanism by which guajadial inhibits tumor growth may be due to multidrug resistance (MDR) by inhibiting the function of P-glycoprotein at the cell membrane level.
