Citrate synthase is an enzyme that catalyzes the condensation of acetyl-CoA and oxaloacetate to form citrate. Citrate synthase is a homodimer composed of two identical subunits, each containing a thiol group. Citrate synthase from porcine heart has been found to be activated by fatty acids, which may be due to its ability to bind to the fatty acid molecule and alter its conformation. Citrate synthase from porcine heart is also used as a chaperone for other proteins and participates in fatty acid synthesis. The citrate synthase enzyme can be cleaved into two fragments: one with an N-terminal domain and the other with the C-terminal domain. These cleavage products can be detected using gel chromatography.
