4-Pentenoic acid, phenylmethyl ester is a potent antiproliferative agent that has been shown to be effective against cancer cells. It is an enantiopure compound with a proton at the 4 position of the phenyl ring and an allyl group at the 3 position. This compound reacts with ethyl bromoacetate in the presence of base to form an aryl halide. The aryl halide can then undergo electrophilic substitution by an aromatic or heteroaromatic halide in the presence of palladium catalyst to give substituted phenols as well as 1,4-dihydropyridines. The lactam moiety can undergo hydrolysis to produce 4-pentenoic acid, which is converted back into 4-pentenoic acid, phenylmethyl ester by treatment with base.
