Sch-202676 hydrobromide is a drug that belongs to the class of allosteric modulators. It has been shown to inhibit the proliferation of pluripotent stem cells and tumor cells in combination with ionomycin, an activator of Ca2+ channels. This drug also sensitizes cancer cells to other pathway inhibitors such as temozolomide, an alkylating agent. Sch-202676 hydrobromide inhibits the production of reactive oxygen species (ROS) by inhibiting nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, which is a key enzyme in ROS production. Sch-202676 hydrobromide has been shown to induce sclerosis in mice by blocking intracellular signaling pathways that activate the transcriptional regulator p38 mitogen-activated protein kinase (MAPK). This leads to hyperactivity and increased cell death in neurons.
