2-((Benzyloxy)methyl)propane-1,3-diol (BPD) is a phosphite that has been used as an intermediate in the synthesis of bisphosphonates. The conversion of BPD to 2-((benzyloxy)methyl)propane-1,3-diol phosphate (BPAP) can be achieved by chloromethylation followed by reduction with triisopropyl phosphite. BPAP can also be obtained by alkylation of 3-hydroxypropionitrile with chloroacetaldehyde and subsequent oxidation. BPAP has been shown to bind to purines and pyrimidines in DNA and inhibit their incorporation into the growing strand. This leads to DNA damage which is repaired by enzymatic repair systems.
