It is a potent and selective inhibitor of phosphodiesterase 1 (PDE1), with an IC50 value of less than 10 nM. MK0893 inhibits PDE1 in vitro and in vivo, leading to increased cAMP levels, vasodilation, and induction of nitric oxide synthase. MK0893 has been shown to have therapeutic efficacy in animal models of systemic sclerosis. In these models, MK0893 inhibited the development of skin fibrosis by inhibiting TGFβ signaling, which is implicated in the pathogenesis of systemic sclerosis.
