5,5-Diphenyl-2-thiohydantoin is a polymerase chain inhibitor that binds to the active site of RNA polymerase, thereby inhibiting transcription and replication. It has been shown to inhibit protein synthesis and to have an antiproliferative effect on cancer cells. The 5,5-Diphenyl-2-thiohydantoin molecule contains a hydroxyl group in the 5 position and two thiol groups in the 2 position. The two thiol groups are important for binding to the polymerase chain. This active form is metabolized through a number of metabolic transformations, including hydrolysis by esterases or glucuronidases, oxidation by cytochrome P450 enzymes, reduction by glutathione reductase, or conjugation with glucuronic acid.
