Ro 64-5229 is a potent and selective agonist of the metabotropic glutamate receptor subtype mGluR2. This drug has been shown to reduce neuroinflammation by inhibiting microglia activation in neonatal rats with hypoxic-ischemic encephalopathy. Ro 64-5229 reduces glutamate release from nerve terminals and its subsequent excitotoxic effects by acting as an allosteric modulator of synaptic transmission. Ro 64-5229 also has the ability to cross the blood brain barrier, which makes it a promising therapy for neuroinflammatory diseases such as Parkinson's disease, multiple sclerosis, and Alzheimer's disease.
A number of studies have demonstrated that Ro 64-5229 can be used to treat certain neurological disorders in newborns, such as neonatal hypoxic-ischemic encephalopathy (NHIE).
