The synthesis of the reaction product is carried out in dichloromethane. The amino acid-carboxylic acid is dissolved in a mixture of dry dichloromethane and tetrafluoroborate at room temperature. The Fmoc-protected glycine, lysine, and arginine are added to the solution dropwise with stirring. After the addition, the reaction system is stirred for 1 h at room temperature and then heated to 50 ˚C for 16 h. The hydrolysis of the ester group is performed with hydrazine in methanol at 0 ˚C.
The resulting product is purified by recrystallization from methanol/water or by preparative HPLC separation.
