5-Aza-2'-deossicitidina, noto anche come decitabina, è un analogo nucleosidico della citidina che viene utilizzato principalmente nel trattamento di alcuni tipi di cancro, in particolare sindromi mielodisplastiche e leucemia mieloide acuta. La sua struttura chimica presenta una base pirimidinica, specificamente una citosina modificata, in cui l'atomo di azoto nella posizione 5 è sostituito da un atomo di carbonio, risultando nell'incorporazione di un gruppo metile. Questa modifica inibisce le metiltransferasi del DNA, portando a ipometilazione del DNA e riattivazione di geni silenziati, il che può indurre differenziazione e apoptosi nelle cellule tumorali. 5-Aza-2'-deossicitidina viene tipicamente somministrato per via endovenosa e ha una vita media relativamente breve, richiedendo protocolli di dosaggio e somministrazione accurati. I suoi effetti terapeutici sono accompagnati da potenziali effetti collaterali, tra cui mielosoppressione e disturbi gastrointestinali. Come strumento di ricerca, è anche utilizzato negli studi epigenetici per indagare il ruolo della metilazione del DNA nell'espressione genica e nella funzione cellulare.
