Acido etacrinico è un composto organico sintetico classificato come diuretico dell'anello, utilizzato principalmente nel trattamento dell'edema associato a insufficienza cardiaca congestizia, cirrosi epatica e malattie renali. È caratterizzato dalla sua formula chimica C13H12ClO4S e da un peso molecolare di circa 299,75 g/mol. Acido etacrinico è una polvere cristallina bianca a bianco sporco che è solubile in acqua e in solventi organici come etanolo e metanolo. Il suo meccanismo d'azione implica l'inibizione del co-trasportatore sodio-potassio-cloruro nei tubuli renali, portando a un aumento dell'escrezione di sodio, cloruro e acqua. Questo effetto diuretico aiuta a ridurre il sovraccarico di liquidi nei pazienti. Acido etacrinico può anche avere effetti collaterali potenziali, inclusi squilibri elettrolitici, disidratazione e ototossicità, in particolare quando usato in alte dosi o in pazienti con condizioni preesistenti. A causa della sua struttura unica, è distinto da altri diuretici dell'anello, come il furosemide, ed è spesso utilizzato in pazienti allergici ai derivati della sulfonamide.

Ethacrynic acid is a non-competitive, reversible inhibitor of the Na+/K+-ATPase. It has been shown to be effective against cancer tissues and infectious diseases such as malaria, tuberculosis, and leishmaniasis. Ethacrynic acid inhibits 2,4-dichlorobenzoic acid (2,4-DCBA)-induced tumor growth in mice by inducing apoptosis in human leukemia cells. Ethacrynic acid also inhibits mitochondrial membrane potential and cellular physiology by decreasing the activity of enzymes which are involved in energy metabolism.
