Acido 4-aminosalicilico, noto anche come acido para-aminosalicilico (PAS), è un composto aromatico con la formula molecolare C7H9NO3. Presenta sia un gruppo amminico (-NH2) che un gruppo acido carbossilico (-COOH) attaccati a una struttura di acido salicilico, rendendolo un membro della famiglia dei salicilati. Questo composto appare come una polvere cristallina bianca o bianco sporco ed è solubile in acqua, il che ne migliora la biodisponibilità. Acido 4-aminosalicilico è principalmente conosciuto per il suo utilizzo come agente antitubercolare, in particolare nel trattamento della tubercolosi, specialmente nei casi resistenti ad altri farmaci. CAS:4-Aminosalicylic acid (4-ASA) is a drug with anti-inflammatory and analgesic properties. It is used in the treatment of bowel disease, including ulcerative colitis and Crohn's disease. 4-ASA binds to the 4th carbon on the para position of p-hydroxybenzoic acid, which is a metabolite of salicylic acid. This binding inhibits the activity of proton pumps that are responsible for maintaining intracellular pH levels in cells. It also prevents the production of inflammatory prostaglandins by inhibiting cyclooxygenase 1 and 2, which are enzymes that catalyze the conversion of arachidonic acid to prostaglandin H2. 4-ASA also has an effect on bacterial infections by interfering with hydrogen bonding interactions between bacterial dna gyrase and dna topoisomerase II, preventing DNA replication.
